T cell exhaustion, i.e. the loss of T cell function and deletion of virus-specific T cell responses, is a typical feature of chronic viral infection. In contrast, infection with cytomegalovirus (CMV), which is evident in 60–80% of the population at 80 years old in the developed world, drives a very different T cell response. CMV-specific T cells adopt a memory-effector phenotype and are maintained or accumulate in a process named ``memory inflation''. On pages 84–90, O’Hara et al., examine the factors that contribute to memory inflation and discuss how CMV-specific T cells might affect immune responses in the elderly. They also consider the possibility of exploiting memory inflation in vaccine strategies to protect against infection and cancer. Cover image designed by Geraldine O’Hara, University of Oxford.
